The wiring diagram decade: what connectomics just proved possible.

To understand a circuit, start with its schematic. For brains, that schematic is the connectome — every neuron and every synapse, mapped. For decades this was dismissed as impossibly hard. Then, in the span of eighteen months, the field delivered two landmarks that changed what "possible" means.

In October 2024, the FlyWire Consortium published the complete wiring diagram of an adult fruit fly brain in Nature: 139,255 neurons connected by roughly 54.5 million chemical synapses — the first whole-brain connectome of an animal that walks, flies, courts, and learns. Six months later, the MICrONS Consortium published a cubic-millimeter reconstruction of mouse visual cortex: 84,035 segmented neurons and 524 million synapses, with recorded activity from around 75,000 of those same neurons. That last part is the revolution — structure and function in the same tissue, letting researchers ask not just what the circuit is, but what it does.

The field's momentum is now institutional. Nature Methods named electron-microscopy connectomics its Method of the Year for 2025, and in February 2026 Princeton's Mala Murthy and Sebastian Seung shared the Wiley Prize in Biomedical Sciences for connectome mapping. Whole-mammalian-brain maps have moved from fantasy to roadmap.

Connectomes, ultimately, are training data. Emerging brain foundation models learn to predict neural activity the way language models predict text — and a synapse-level wiring diagram is the ground truth that keeps those models honest. The fly proved the pipeline. The mouse cube proved it scales. The question the field now organizes around is simple: what needs to be built so that, one day, the same can be said of a human brain — ethically and openly.
